The GSure® Sputum DNA Isolation Kit is designed to efficiently extract high-quality genomic DNA from freshly collected sputum samples, including thick, mucous-rich, and clinically processed specimens. Sputum contains viscous mucopolysaccharides, cell debris, protein aggregates, and PCR inhibitors that make extraction challenging. The GSure® system overcomes these obstacles using a three-buffer lysis cascade (GDSP1 → GDSP2 → GDSP3) optimized for viscosity reduction, mucolytic action, and effective DNA release.
Purified DNA is suitable for PCR, qPCR, sequencing, genotyping, and molecular diagnostic assay development—especially in TB, respiratory pathogen, and clinical microbiology workflows.

The GSure® Sputum DNA Isolation Kit uses a multi-stage lysis system tailored for viscous sputum, followed by silica-column purification.
● Sample Lysis Add 250 µL GDSP1 to 200–300 µL sputum → incubate at 70°C for 15 minutes, vortex every 2 minutes.
For swab samples: immerse swab in 300 µL GDSP1, roll and press against tube wall.
Add 250 µL GDSP2, invert gently and incubate again at 70°C for 15 minutes.
Add 350 µL GDSP3, mix by inversion (no vortexing).
Centrifuge 10 minutes @ 13,000 rpm to pellet debris.
● Binding to Column Transfer supernatant carefully to GMini Spin Column (avoid pellet).
Spin at 13,000 rpm for 30–60 seconds and discard flow-through.
● Washing Add 600 µL Membrane Wash Buffer and spin.
Repeat wash.
Perform an additional 2-minute dry spin to remove ethanol—critical for enzymatic reactions.
● Elution Add 50 µL Nuclease-Free Water to center of the membrane.
Stand for 1 minute.
Spin 1 minute @ 13,000 rpm to elute purified DNA
