How the Assay Works
The BCA assay is based on the reduction of Cu²⁺ to Cu⁺ by peptide bonds and specific amino acid residues (cysteine, cystine, tryptophan, tyrosine) under alkaline conditions. Bicinchoninic acid (BCA), a water-soluble chromogenic reagent, chelates Cu⁺ to form a purple complex with strong absorbance at 562 nm. The intensity of this color is directly proportional to the protein concentration in the sample.
