Intent to use:
The DiAGSure® JAK2 V617F Mutation Detection Kit is a highly sensitive real-time PCR assay designed for the qualitative detection of the V617F point mutation in the JAK2 gene. This mutation, a guanine-to-thymine substitution at nucleotide 1849, results in a valine-to-phenylalanine substitution and is found in approximately 95% of patients with polycythemia vera (PV), and in 50–60% of patients with essential thrombocythemia (ET) and primary myelofibrosis (PMF). The mutation leads to constitutive activation of the JAK-STAT signaling pathway, promoting uncontrolled myeloproliferation. Early detection is critical for accurate diagnosis, classification, and therapeutic monitoring of myeloproliferative neoplasms (MPNs). The assay delivers high specificity and sensitivity, even in samples with low mutant allele burden, supporting risk stratification and treatment decisions. It is an essential molecular tool for hematologists managing patients with suspected or confirmed MPNs.
