The GSure® Plasmid DNA Isolation Kit provides a robust alkaline lysis workflow followed by silica-column purification to recover highly pure plasmid DNA.
● Resuspension
Complete dispersal of the bacterial pellet using GSPB1 ensures uniform lysis. Avoid visible clumps to prevent genomic DNA contamination. (GSPB1 must be stored at 4°C.)
● Alkaline Lysis + Sheer PINK Indicator
Addition of GSPB2 initiates lysis, turning the solution pink. Gentle inversion (no vortexing) ensures controlled lysis. A clear pink solution indicates complete lysis.
● Neutralization
GSPB3 is added, turning the mixture colorless and cloudy, marking complete neutralization and precipitation of cell debris, genomic DNA, and proteins.
● Clarification
Centrifugation pellets debris; the cleared lysate is carefully loaded onto the GMini column.
● DNA Binding and Washing
The plasmid binds to the silica membrane; subsequent wash steps with ethanol-enriched Membrane Wash Buffer ensure removal of salts and contaminants.
● Elution
Plasmid DNA is eluted using 50 µL nuclease-free water, producing clean, ready-to-use DNA for molecular workflows.
