Trizine™ uses a proven monophasic solution of phenol and guanidinium thiocyanate to simultaneously lyse cells, inactivate RNases, and preserve RNA. After sample homogenization, the addition of chloroform induces phase separation, allowing RNA to partition into the upper aqueous phase while DNA and proteins separate into the interphase and organic layer.
RNA is then recovered by isopropanol precipitation, washed with ethanol, and resuspended in nuclease-free water. The resulting RNA is free of protein, DNA, polysaccharides, and enzymatic inhibitors.
This chemistry provides excellent RNA stability, even from RNase-rich tissues, ensuring highly reproducible gene expression data.
