The workflow incorporates:
● Sample Lysis (Pre-warmed WLN + PB + RW1 mix, Proteinase K)
Tissue is crushed (with or without liquid nitrogen), treated with the preheated buffer mixture, and incubated at 65°C for enhanced breakdown of plant cell walls.
● Inhibitor Removal (AMD Buffer)
AMD Buffer precipitates pigments, polysaccharides, and polyphenols, ensuring maximal inhibitor removal and clear lysate formation.
● DNA Binding Using RP1 Buffer
The clarified lysate is mixed with RP1 Binding Buffer and loaded onto the GMini DNA Binding Column.
● Column Purification
Washing with Membrane Wash Buffer removes residual contaminants.
