GSure Plasmid DNA Isolation Kit is an alkaline lysis–based system designed for rapid purification of covalently closed circular (ccc) plasmid DNA from fresh overnight E. coli cultures. The kit uses a silica-membrane spin column and three optimized buffers to ensure complete cell lysis, efficient protein removal, and highly purified plasmid recovery within 30 minutes. A unique chromogenic indicator, SheerPink, visually confirms proper lysis and neutralization, minimizing genomic DNA carryover and ensuring maximum ccc plasmid integrity.
The kit isolates plasmids of varying sizes with equal efficiency, successfully recovering constructs from 2.6 kb to 18 kb. It yields approximately ≥3 μg of plasmid DNA from 1 ml of high-copy cultures, while low-copy plasmids typically yield 500 ng–1.5 μg per ml. Sample volumes from 1–12 ml can be processed, although the highest proportion (>75%) of ccc DNA is obtained from ≤3 ml culture; larger volumes do not reduce total yield but slightly lower ccc percentage.
Spectrophotometric analysis shows consistently clean preparations, with A260/A280 ratios of 1.79–1.83, indicating minimal protein or RNA contamination. PCR using 16S rDNA primers produces no amplification, confirming the absence of trace genomic DNA. The resulting plasmid DNA is compatible with all major downstream applications, including restriction digestion, PCR, Sanger sequencing, and in vitro transcription, demonstrating the kit’s reliability for routine molecular biology workflows.
